SPHR980 is an orally bioavailable small molecule inhibitor specifically targeting Aurora kinase A (AURKA). Developed by Sphaera Pharma, SPHR980 is designed to disrupt the function of Aurora A, a serine/threonine kinase that plays a critical role in mitosis, particularly in centrosome maturation and spindle assembly. Overexpression of Aurora A is associated with chromosomal instability and poor prognosis in various malignancies, including breast, colorectal, and pancreatic cancers. By inhibiting Aurora A, SPHR980 induces mitotic arrest and apoptosis in cancer cells. It has demonstrated significant anti-tumor activity in preclinical models and has been investigated for the treatment of advanced solid tumors.
