Drug intelligence / Profile preview
**SPI-77** was an investigational long-circulating, PEGylated, sterically stabilized liposomal formulation of cisplatin developed by ALZA Pharmaceuticals, formerly SEQUUS Pharmaceuticals. The liposomal carrier was intended to reduce peak free-cisplatin exposure and preferentially accumulate in tumors through enhanced permeability and retention, while released cisplatin forms covalent DNA adducts and crosslinks that impair DNA replication and transcription and promote tumor-cell death. Clinical studies in non-small-cell lung cancer and platinum-sensitive recurrent ovarian cancer showed limited antitumor activity; development was discontinued, in part because the formulation released insufficient cisplatin at tumor sites.
