Overview

SPI-77 + vinorelbine is a combination chemotherapy regimen consisting of SPI-77, a liposomal pegylated formulation of cisplatin (marketed as STEALTH cisplatin), and the vinca alkaloid chemotherapeutic agent vinorelbine. SPI-77 was developed to reduce the systemic toxicity associated with conventional cisplatin by encapsulating it in liposomes, thereby improving tumor targeting and minimizing nephrotoxicity and neuropathy. Vinorelbine disrupts microtubule formation, inhibiting mitosis in cancer cells. This combination has been investigated primarily for advanced non-small cell lung cancer (NSCLC), particularly in patients refractory to or recurrent following previous chemotherapy[1][2][4]. The regimen was generally well tolerated; neutropenia was identified as the dose-limiting toxicity at higher doses of SPI-77[1]. Partial responses were observed in some patients.
