Spike Ferritin Nanoparticle (SpFN) is a protein-based vaccine candidate designed to provide broad protection against SARS-CoV-2 and related sarbecoviruses. Developed primarily by the Walter Reed Army Institute of Research (WRAIR), the vaccine utilizes a self-assembling ferritin nanoparticle scaffold to display 24 subunits of the SARS-CoV-2 spike protein in an organized, repetitive array. This multivalent presentation mimics the structure of a virus, enhancing the activation of B-cells and the production of neutralizing antibodies. The vaccine is typically adjuvanted with the Army Liposomal Formulation (ALFQ), which contains monophosphoryl lipid A and QS-21, to stimulate robust Th1-biased cellular immune responses. Another version, DCFHP (Developing Countries Federated Health Partnership), uses a similar ferritin-based architecture and has been evaluated with aluminum-salt adjuvants.
