Drug intelligence / Profile preview
SPIONs-DSS6 is a bone-targeted superparamagnetic iron oxide nanoparticle (SPION) platform conjugated with the bone-homing peptide DSS6 (six repetitive sequences of aspartate, serine, and serine). Developed by researchers at Nanjing University and Nanjing Drum Tower Hospital, SPIONs-DSS6 is designed to treat senile osteoporosis (SOP) by mimicking exercise-induced mechanical stimulation. When combined with an external variable magnetic field (VMF), SPIONs-DSS6 targets osteocytes and generates localized mechanical forces. This mechanical stimulation activates PGC-1α-dependent mitophagy, which restores mitochondrial homeostasis, reduces oxidative stress, limits mitochondrial DNA leakage, and suppresses osteocyte senescence, ultimately improving bone mass and mitigating osteoporosis progression.
