OBP-801 (spiruchostatin A) is a small molecule histone deacetylase (HDAC) inhibitor with potent and specific activity against HDAC enzymes. By inhibiting HDACs, OBP-801 leads to the accumulation of acetylated histones, chromatin remodeling, and altered gene expression patterns that promote the transcription of tumor suppressor genes. This results in inhibition of tumor cell division and induction of apoptotic and autophagic cell death in cancer cells. Preclinical studies have shown that OBP-801 has more potent HDAC inhibitory activity than other approved HDAC inhibitors such as vorinostat (Zolinza) and romidepsin (Istodax), with efficacy expected across a wide range of cancers. It has also been investigated for potential ophthalmic uses in collaboration with Kyoto Prefectural University of Medicine. Target indications include lung cancer, lymphoma, kidney cancer (including clear cell renal cell carcinoma), glaucoma, solid tumors generally, and muscle-invasive bladder cancer[1][2][5][6][8].
