SPL-108 is an investigational 8-amino acid peptide drug developed by Splash Pharmaceuticals that targets the cancer stem cell antigen CD44. It acts as a CD44 inhibitor, modulating CD44-mediated intracellular signaling and reducing multidrug resistance, particularly to paclitaxel and platinum agents in cancer cells. Preclinical studies have shown that SPL-108 possesses antiangiogenic, antimigratory, anti-invasive, and antimetastatic properties against various cancer cell lines. Its mechanism involves binding to CD44 (the main ligand of hyaluronic acid), thereby affecting P-glycoprotein (MDR1) function and potentially increasing sensitivity to chemotherapeutic agents. Clinical trials have primarily focused on platinum-resistant ovarian, primary peritoneal, or fallopian tube cancers expressing CD44; however, preclinical data suggest potential utility in other tumor types such as breast and endometrial cancers[1][2][3][5][6].
