Overview

SPL-334.1 (also known as GTI-334.1) is a preclinical-stage small molecule inhibitor of S-nitrosoglutathione reductase (GSNOR), developed by GSNOR Therapeutics. GSNOR is a key enzyme involved in the metabolism of S-nitrosoglutathione (GSNO), which serves as a major reservoir for nitric oxide (NO) bioactivity. By inhibiting GSNOR, SPL-334.1 increases the concentration of GSNO, leading to enhanced NO signaling that promotes bronchodilation, reduces inflammation, and inhibits fibrotic processes. The drug is being investigated for multiple indications, including idiopathic pulmonary fibrosis (IPF), asthma, systemic inflammation, myocardial infarction (heart attack), and non-alcoholic steatohepatitis (NASH). Preclinical studies have utilized oral, intravenous, and intraperitoneal routes of administration.
