Overview

SPL-891.1 (also known as GTI-891.1) is a small molecule inhibitor of S-nitrosoglutathione reductase (GSNOR) being developed by GSNOR Therapeutics. GSNOR is an enzyme that regulates the levels of S-nitrosoglutathione (GSNO), a key signaling molecule in nitric oxide (NO) biology. By inhibiting GSNOR, SPL-891.1 aims to increase GSNO levels, thereby enhancing S-nitrosylation and modulating various inflammatory and fibrotic pathways. The drug is currently in preclinical development for a wide range of indications, including idiopathic pulmonary fibrosis (IPF), non-alcoholic steatohepatitis (NASH), rheumatoid arthritis, diabetes, Crohn's disease, heart failure, stroke, and neurodegenerative disorders. It has been studied across multiple routes of administration, including oral, intravenous, intraperitoneal, and intravitreal.
