**SPL026 + selective serotonin reuptake inhibitor** is a combination therapy consisting of SPL026 (a proprietary, pharmaceutical-grade formulation of intravenous N,N-dimethyltryptamine, or DMT) administered together with a selective serotonin reuptake inhibitor (SSRI), a class of antidepressants. SPL026 acts as a psychedelic agonist primarily at the serotonin 2A receptor, inducing rapid and intense psychedelic experiences. SSRIs block reuptake of serotonin via the serotonin transporter, leading to increased synaptic serotonin levels and gradual neurochemical changes. Recent clinical studies in patients with major depressive disorder (MDD) show that co-administration of SPL026 and SSRIs is safe and well-tolerated, with evidence of enhanced antidepressant efficacy compared to SPL026 alone.
