Overview

SPM001 is a first-in-class stapled peptide inhibitor developed by SPIMA Therapeutics. It specifically targets the Myddosome complex, specifically the MyD88 (Myeloid differentiation primary response 88) protein. MyD88 is a critical adapter protein in the Toll-like receptor (TLR) and Interleukin-1 receptor (IL-1R) signaling pathways. Mutations in MyD88, such as the L265P mutation, lead to constitutive activation of the Myddosome, driving the progression of various B-cell malignancies and inflammatory conditions. By disrupting the assembly or function of the Myddosome complex, SPM001 aims to inhibit downstream NF-κB signaling. It is currently in preclinical development for the treatment of MyD88-mutated cancers and inflammatory disorders.
