SPmAb is a proprietary monoclonal antibody platform and a series of therapeutic candidates developed by Vaxil BioTherapeutics. It represents a novel class of antibodies that specifically target the signal peptide (SP) domain of tumor-associated antigens, with the lead candidates SPmAb-2.1 and SPmAb-6 targeting the MUC1 signal peptide. Unlike conventional anti-MUC1 antibodies that target the extracellular tandem repeat domain—which is often shed into the circulation and can act as a decoy—SPmAbs target the MUC1 signal peptide that is uniquely presented on the surface of malignant cells. This targeting mechanism induces potent complement-dependent cytotoxicity (CDC) against MUC1-expressing cancer cells, including those in multiple myeloma and various solid tumors. Additionally, SPmAb is being developed as a companion diagnostic to identify patients likely to benefit from Vaxil's ImMucin vaccine therapy.
