Overview

SPN-821 is an investigational, orally bioavailable small molecule being developed by Supernus Pharmaceuticals for the treatment of major depressive disorder (MDD). It functions as a first-in-class intracellular modulator of the mechanistic target of rapamycin complex 1 (mTORC1), a protein complex that plays a critical role in synaptic plasticity, cellular metabolism, and protein synthesis in the brain. By directly enhancing mTORC1 signaling, SPN-821 is designed to increase synaptic function and connectivity, potentially providing rapid-onset antidepressant effects without the dissociative side effects associated with NMDA receptor antagonists like ketamine. The compound is currently being evaluated in Phase 2 clinical trials as an adjunctive therapy to standard antidepressants, administered at a dose of 2400 mg twice weekly.
