SPOT-03 is an investigational non-viral gene therapy developed by Spot BioSystems for the treatment of Duchenne muscular dystrophy (DMD). It utilizes a proprietary extracellular vesicle (EV)-mediated delivery platform to transport mRNA encoding a functional version of the dystrophin protein directly to muscle cells. Unlike traditional viral vector-based gene therapies, SPOT-03's EV-mediated approach is designed to be non-immunogenic, potentially allowing for repeat administration and avoiding the cargo size limitations associated with adeno-associated virus (AAV) vectors. The program is currently being evaluated in a Phase 1 first-in-human clinical trial.
