**SPP 635** is a small molecule, non-peptide **renin inhibitor** developed by Speedel (later acquired by Novartis) and Speedel Pharma AG for the treatment of **hypertension**, **albuminuria**, **type 2 diabetes mellitus with nephropathy**, and for **end-organ protection**. The drug functions by inhibiting renin, an enzyme that catalyzes the first step in the renin–angiotensin–aldosterone system (RAAS), thereby reducing the production of angiotensin I and ultimately lowering blood pressure. SPP 635 demonstrated a dose-dependent reduction in intraocular pressure (IOP) when topically administered to glaucomatous monkeys, suggesting it also has potential in lowering IOP for glaucoma. Clinical trials were conducted to assess its safety and efficacy, particularly in mild to moderate hypertension with albuminuria and diabetic nephropathy. Despite initial promise, clinical development was discontinued after phase II trials.
