Drug intelligence / Profile preview
SPP1 monoclonal antibody (also known as anti-osteopontin or anti-OPN monoclonal antibody) is a class of targeted biologic therapies designed to neutralize secretory phosphoprotein 1 (SPP1), a multifunctional extracellular matrix glycoprotein and cytokine also known as osteopontin (OPN). SPP1 is highly expressed by tumor-associated macrophages (TAMs), degenerative renal tubular epithelial cells, and other immune and stromal cells in various disease states. By binding to SPP1, these antibodies disrupt its interaction with cell-surface receptors such as CD44 and various integrins (e.g., αvβ3, α9β1), thereby inhibiting downstream pro-inflammatory, pro-fibrotic, and immunosuppressive signaling pathways (such as PI3K/AKT and MAPK). Preclinical and clinical studies have investigated SPP1 monoclonal antibodies for the treatment of solid tumors (including hepatocellular carcinoma, colorectal cancer liver metastasis, and breast cancer), rheumatoid arthritis, and chronic kidney disease/renal fibrosis. Notable specific candidates in this class include ASK8007 (originally developed by Astellas Pharma and KAKETSUKEN) and AOM1 (developed by Pfizer).
