Overview

SPP1-neutralizing antibody is a biologic therapeutic candidate designed to inhibit the activity of Secreted Phosphoprotein 1 (SPP1), also known as osteopontin. SPP1 is a multifunctional glycoprotein that acts as a key mediator in the pathogenesis of idiopathic pulmonary fibrosis (IPF), where it is highly expressed by monocyte-derived macrophages. By binding to and neutralizing SPP1, the antibody disrupts profibrotic signaling pathways and reduces the expression of fibroproliferative markers such as CTHRC1. Preclinical studies conducted by researchers at Baylor College of Medicine and other institutions have demonstrated that neutralizing SPP1 can attenuate fibrotic progression in both ex vivo precision-cut lung slices and in vivo bleomycin-induced injury models, suggesting that targeting the SPP1 pathway is a viable therapeutic strategy for treating pulmonary fibrosis.
