Overview

SPP1100 is an experimental small molecule direct renin inhibitor (DRI) that was under development by Speedel for the treatment of hypertension. As a member of the DRI class, SPP1100 functions by binding to the active site of the enzyme renin, which catalyzes the rate-limiting step in the renin-angiotensin-aldosterone system (RAAS). By inhibiting renin, the drug prevents the conversion of angiotensinogen to angiotensin I, ultimately reducing the production of the potent vasoconstrictor angiotensin II. This mechanism leads to systemic vasodilation and a reduction in blood pressure. SPP1100 was part of a broader portfolio of renin inhibitors at Speedel, which included the first-in-class agent aliskiren (SPP100). Following the acquisition of Speedel by Novartis in 2008, the development of SPP1100 was terminated as the company focused on its lead commercial and late-stage assets.
