SPP630 is a potent, orally active, second-generation small molecule renin inhibitor originally developed by the Swiss biopharmaceutical company Speedel. It was designed to target the renin-angiotensin-aldosterone system (RAAS) by inhibiting renin, the enzyme responsible for the rate-limiting step in the production of angiotensin II. By blocking the conversion of angiotensinogen to angiotensin I, SPP630 was intended to treat hypertension and provide protective effects against cardiovascular and renal damage. Following the acquisition of Speedel by Novartis in 2008, the development of several pipeline renin inhibitors, including SPP630, was evaluated against the existing commercial success of aliskiren (the first-in-class renin inhibitor). SPP630 was engineered for improved potency and pharmacokinetic properties compared to first-generation candidates.
