SPP676 is an orally active, small-molecule renin inhibitor that was developed for the treatment of hypertension and related cardiovascular diseases. It belongs to a class of drugs designed to block the renin-angiotensin-aldosterone system (RAAS) at its rate-limiting step—the conversion of angiotensinogen to angiotensin I by the enzyme renin. Developed by the Swiss biopharmaceutical company Speedel, SPP676 was part of a pipeline of next-generation renin inhibitors intended to offer improved potency or pharmacokinetic profiles compared to the first-in-class inhibitor, aliskiren. Following the acquisition of Speedel by Novartis in 2008, development of several early-stage pipeline candidates, including SPP676, was deprioritized or discontinued in favor of established products like aliskiren (Tekturna).
