Overview

SPR719 is a novel small molecule aminobenzimidazole antibacterial agent that acts as the active moiety of the oral prodrug SPR720. It functions primarily by inhibiting the ATPase subunit of bacterial DNA gyrase B (GyrB), a target not exploited by current antibiotics, and also inhibits DNA topoisomerase IV. This dual mechanism disrupts bacterial DNA replication and transcription, leading to bactericidal activity. SPR719 demonstrates potent in vitro activity against clinically relevant mycobacterial species including Mycobacterium avium complex (MAC), Mycobacterium abscessus, Mycobacterium kansasii, and Mycobacterium tuberculosis—including drug-resistant strains. The drug has shown low propensity for resistance development and additive effects when combined with standard-of-care agents such as clarithromycin and ethambutol. Spero Therapeutics is developing this agent for nontuberculous mycobacterial pulmonary disease (NTM-PD) and tuberculosis[1][2][3][5][6][7].
