Sprint Bioscience-TREX1 is a preclinical immuno-oncology program consisting of small molecule inhibitors that target the three-prime repair exonuclease 1 (TREX1) protein. Originally developed by Sprint Bioscience and acquired by Gilead Sciences in November 2025, these inhibitors are designed to prevent the degradation of cytosolic DNA fragments within cancer cells. By blocking TREX1, the program aims to activate innate immune signaling pathways and boost type I interferon responses, which enhances the immune system's ability to detect and attack tumors. Preclinical studies in animal models of colorectal cancer have demonstrated that oral administration of these inhibitors can reduce tumor growth and improve immune responses, suggesting potential for use as a monotherapy or in combination with existing immunotherapies, radiotherapy, and chemotherapy.
