SPT-310 is an oral prodrug of cannabidiol (CBD) developed by Seaport Therapeutics (formerly part of PureTech Health) using the proprietary Glyph™ platform. The Glyph technology utilizes a lipid-conjugation strategy to direct the drug's absorption into the lymphatic system, bypassing the portal vein and first-pass hepatic metabolism. This approach is intended to enhance the oral bioavailability of CBD and reduce the potential for hepatotoxicity and gastrointestinal side effects typically associated with high-dose, oil-based CBD formulations. Preclinical data in rodent models of maximal electroshock seizures (MES) indicate that SPT-310 provides potent anti-convulsant activity with a significantly lower CBD-equivalent ED50 compared to standard oral CBD, suggesting potential for improved therapeutic efficacy and safety in patients with epilepsy, including Dravet syndrome and Lennox-Gastaut syndrome.
