Overview

SPT-348 is a non-hallucinogenic psychoplastogen and serotonin receptor modulator of the lysergamide family, currently under development for the treatment of depression, anxiety, and other neuropsychiatric disorders. It acts as a prodrug of 2-bromo-LSD (bromolysergide; BOL-148), which is a non-selective serotonin receptor modulator and partial agonist at the serotonin 5-HT2A receptor. Unlike LSD, SPT-348 does not induce hallucinogenic effects. The drug utilizes Seaport Therapeutics' proprietary Glyph platform—a triglyceride linker formulation designed to bypass first-pass hepatic metabolism—improving pharmacokinetics and tolerability compared to conventional psychedelics. As of 2025, SPT-348 remains in preclinical or discovery-stage development[1][4][5][7].
