SPU-21 is a synthetic peptide drug developed as a liposomal joint homing agent that selectively targets inflamed synovial tissue, inhibiting progression of rheumatoid arthritis (RA)[1][3][5][8]. The drug functions by identifying markers of arthritic inflammation, allowing for localized delivery of anti-arthritic therapeutics, such as dexamethasone, with potentially enhanced efficacy over systemic administration[2][3][5]. SPU-21 is designed for peptide-guided delivery and may be customizable for use with other agents including nanoparticles, fusion imaging molecules, or targeted delivery of therapeutics such as psilocybin[3][5][6]. Developed in collaboration between Silo Pharma and the University of Maryland, Baltimore, it is currently being investigated for treatment of autoimmune diseases, primarily rheumatoid arthritis, with emphasis on subcutaneous administration for patient convenience and efficacy[2][4][5].
