Overview

SPX-0152 is an oral, once-daily small molecule modulator of α4β2 nicotinic acetylcholine receptors (nAChRs) being developed by SpaceRx for smoking cessation and nicotine addiction. The drug targets the α4β2 nAChR subtype, which is the principal mediator of nicotine's reinforcing effects in the brain. SpaceRx employs a proprietary platform involving in-vivo real-time biosensors to evaluate and optimize prodrug candidates designed to provide sustained therapeutic levels and prolong therapeutic effects. The program has received National Institutes of Health (NIH) Small Business Innovation Research (SBIR) funding and is currently in the IND-enabling stage, with Phase 1 clinical trials expected to commence in 2026.
