Overview

SPX-304 is a preclinical bispecific monoclonal antibody developed by SparX Biotech. It is engineered to simultaneously target Programmed cell death 1 ligand 1 (PD-L1) and Leukocyte surface antigen CD47 (CD47). The dual-targeting mechanism is designed to enhance anti-tumor immunity by blocking the PD-1/PD-L1 checkpoint axis to restore T-cell activity while concurrently inhibiting the CD47/SIRPα "don't eat me" signaling pathway to promote macrophage-mediated phagocytosis of cancer cells. SPX-304 was identified through SparX's proprietary SAILING platform, which utilizes AI-driven optimization for the discovery of multi-specific antibodies. As of 2024, the program remains in the early discovery or preclinical stage, primarily intended for the treatment of solid tumors.
