SPX-308 is a biparatopic bispecific antibody currently in preclinical development by SparX Biopharmaceutical for the treatment of cancer. It is designed to simultaneously target Vascular Endothelial Growth Factor (VEGF) and Programmed Death-Ligand 1 (PD-L1). By inhibiting VEGF, SPX-308 aims to disrupt tumor angiogenesis and normalize the tumor vasculature, while its PD-L1 targeting component works to block the PD-1/PD-L1 immune checkpoint, thereby restoring the anti-tumor activity of T-cells. The 'biparatopic' nature of the antibody suggests it may bind to two distinct epitopes on one or both of its targets, potentially leading to enhanced binding affinity and therapeutic efficacy compared to standard bispecific antibodies. This dual-targeting approach is intended to provide a synergistic effect by addressing both the vascular and immune-suppressive components of the tumor microenvironment.
