Overview

SPX-603 is a tri-specific monoclonal antibody developed by SparX Biotech, currently in preclinical development for the treatment of solid tumors. Engineered using the company's proprietary SAILING™ platform, SPX-603 is designed to simultaneously target three key drivers of tumor progression and resistance: the epidermal growth factor receptor (EGFR), the hepatocyte growth factor receptor (c-Met/MET), and the human epidermal growth factor receptor 3 (HER3/ErbB3). By inhibiting these three receptor tyrosine kinases, SPX-603 aims to provide a more robust blockade of downstream signaling pathways (such as PI3K/AKT and MAPK) than current mono- or bispecific therapies, potentially overcoming resistance mechanisms like c-Met amplification or HER3 upregulation that often occur following EGFR inhibition. The drug is intended to address advanced malignancies where multi-pathway activation limits the efficacy of standard targeted treatments.
