SPX-629 is a next-generation antibody-drug conjugate (ADC) developed by SparX Biotech that targets the Human Epidermal Growth Factor Receptor 2 (HER2). It utilizes a proprietary dual-payload platform (Nab2) to deliver two distinct cytotoxic agents: a Topoisomerase 1 (TOP1) inhibitor and Monomethyl auristatin E (MMAE). By combining these two mechanisms of action—DNA damage and microtubule inhibition—SPX-629 is designed to enhance anti-tumor activity and overcome potential resistance mechanisms associated with single-payload ADCs. The candidate is currently in the IND-enabling stage of development for the treatment of HER2-expressing solid tumors.
