Overview

SPY002-079 is a novel, extended half-life monoclonal antibody that targets Tumor Necrosis Factor-like Ligand 1A (TL1A), a proinflammatory cytokine in the TNF superfamily. Developed by Spyre Therapeutics, SPY002-079 is designed to inhibit TL1A signaling through the Death Receptor 3 (DR3), a pathway implicated in the pathogenesis of immune-mediated inflammatory diseases such as ulcerative colitis and Crohn's disease. The antibody incorporates specific Fc modifications that attenuate effector functions and enhance binding to the neonatal Fc receptor (FcRn) at acidic pH, significantly extending its pharmacokinetic profile. Preclinical data suggest that these modifications support infrequent dosing schedules, potentially allowing for quarterly or biannual administration. SPY002-079 has demonstrated high-affinity binding to human TL1A and potent functional inhibition in various preclinical models of inflammatory bowel disease and rheumatic diseases.
