SPY002-080 (also referred to as SPY002) is a novel, extended half-life monoclonal antibody that targets tumor necrosis factor-like ligand 1A (TL1A), a proinflammatory cytokine in the TNF superfamily. Developed by Spyre Therapeutics, SPY002 is designed to inhibit TL1A signaling through the death receptor 3 (DR3), a pathway that plays a critical role in the pathogenesis of immune-mediated inflammatory diseases by activating T helper cells, innate lymphoid cells, and fibroblasts. The antibody is engineered with specific Fc modifications to enhance its binding to the neonatal Fc receptor (FcRn) at acidic pH, which significantly extends its terminal half-life. This pharmacokinetic profile is intended to support infrequent dosing schedules, such as quarterly or biannual administration, potentially offering improved patient convenience over first-generation TL1A inhibitors. Preclinical characterization has demonstrated high-affinity binding (KD ≈ 31-35 pM) and potent functional inhibition, with efficacy shown in models of colitis, arthritis, and psoriasis. SPY002 is currently being developed for the treatment of inflammatory bowel diseases, including ulcerative colitis and Crohn's disease, as well as other rheumatic conditions.
