**SPY072** is an investigational, extended half-life monoclonal antibody engineered to target and inhibit TL1A (tumor necrosis factor ligand superfamily member 15, also known as VEGI), a key cytokine implicated in the pathogenesis of several immune-mediated and rheumatologic diseases. Developed by Spyre Therapeutics, it is designed as a next-generation anti-TL1A therapy with significantly prolonged pharmacokinetics—demonstrating a half-life of approximately 75 days, allowing for potential quarterly or less frequent dosing. In Phase 1 studies, single doses up to 1500 mg were well tolerated with no serious adverse events, and the drug exhibited strong, durable suppression of free TL1A levels for at least 20 weeks at the lowest tested dose. SPY072 is being advanced as a monotherapy or as part of combination regimens for immune-mediated diseases, notably rheumatoid arthritis (RA), psoriatic arthritis (PsA), and axial spondyloarthritis (axSpA). Phase 2 trials in those indications (SKYWAY-RD basket trial) are planned for initiation in Q3 2025[1][3][4][6][7].
