SQ-22031 is a recombinant human nerve growth factor (rhNGF) ophthalmic solution developed by Shenyang Xingqi Eye Medicine for the treatment of neurotrophic keratitis (NK). NK is a rare, degenerative corneal disease resulting from the loss of corneal sensory innervation, which leads to epithelial breakdown, impaired healing, and potential corneal perforation. SQ-22031 functions by mimicking the biological activity of endogenous nerve growth factor (NGF), a protein essential for the survival and maintenance of corneal nerves and epithelial cells. By binding to the high-affinity Tropomyosin receptor kinase A (TrkA) and the low-affinity p75 neurotrophin receptor (p75NTR) on the ocular surface, SQ-22031 stimulates corneal nerve regeneration, enhances tear secretion, and promotes the proliferation and differentiation of corneal epithelial cells. The drug is currently in Phase 3 clinical development in China, aiming to provide a therapeutic option for restoring corneal integrity and sensitivity.
