Overview

**SQ2201** is a preclinical, HER2-targeted CAPAC cancer-therapy regimen developed by Shasqi. It combines SQT01, a systemically administered tetrazine-modified HER2-binding antibody fragment, with SQP22, a trans-cyclooctene-modified monomethyl auristatin E protodrug. After SQT01 accumulates at HER2-positive tumor tissue, subsequent SQP22 administration undergoes a bioorthogonal tetrazine-trans-cyclooctene click reaction that releases active MMAE locally, aiming to concentrate microtubule-disrupting cytotoxic activity at the tumor while limiting systemic exposure. Preclinical studies demonstrated antitumor activity in HER2-positive tumor models.
