SQ3370 is a novel investigational cancer therapeutic and the first clinical example of a click chemistry-activated drug delivery system. It is based on the Click Activated Protodrugs Against Cancer (CAPAC) platform and consists of two components: - SQL70, a tetrazine-modified biopolymer injected locally into the tumor - SQP33, an attenuated protodrug form of doxorubicin administered intravenously The two components react via click chemistry at the tumor site to release active doxorubicin directly within the tumor microenvironment. This approach enables much higher local concentrations of doxorubicin with reduced systemic toxicity compared to conventional administration. Preclinical and clinical studies have shown that SQ3370 can be safely administered at doses up to 12 times higher than standard doxorubicin regimens without dose-limiting toxicities or significant cardiotoxicity. The therapy also induces immune activation in tumors, suggesting potential synergy with immunotherapies[1][2][4][7].
