Overview

SQOR shRNA is a gene-silencing therapeutic candidate designed to target sulfide quinone oxidoreductase (SQOR), a rate-limiting enzyme in mitochondrial sulfide oxidation. In glioblastoma (GBM), SQOR and persulfide dioxygenase (ETHE1) are often upregulated and correlate with poor prognosis. By knocking down SQOR, the drug causes an accumulation of hydrogen sulfide (H2S), which inhibits the electron transport chain at complex IV and disrupts mitochondrial respiration and ATP production. This metabolic shift forces cells into a glycolytic state and impairs lipid mobilization, leading to decreased self-renewal and proliferation of glioblastoma stem cells. Research conducted at the Cleveland Clinic has demonstrated that SQOR knockdown significantly extends survival in murine glioblastoma models.
