**SQZ-AAC-HPV + nivolumab** is a combination immunotherapy under investigation for the treatment of HPV16-positive advanced or metastatic solid tumors. The regimen consists of SQZ-AAC-HPV, an engineered cell therapy product based on red blood cells (RBCs) modified using the Cell Squeeze® technology to carry HPV16-specific antigens (E6 and E7 oncoproteins). These engineered RBCs (Activating Antigen Carriers, AACs) are designed to mimic aged RBCs, which are naturally targeted for clearance by antigen-presenting cells (APCs), leading to endogenous CD8+ T cell priming for antitumor responses. Nivolumab is a monoclonal antibody that functions as an immune checkpoint inhibitor, specifically blocking programmed death-1 (PD-1) on T cells to enhance their activity against tumors. Together, these therapies aim to synergistically provoke targeted antitumor immunity by both priming new T cell responses and relieving immune inhibition.
