Overview

This is a combination investigational therapy consisting of SQZ-AAC-HPV, nivolumab, and ipilimumab. **SQZ-AAC-HPV** is an engineered, cell-based immunotherapy developed using SQZ Biotechnologies’ Cell Squeeze technology; it creates Activating Antigen Carriers (AACs) derived from red blood cells that are loaded with HPV16 antigens (E6 and E7 oncoproteins) and aimed at priming the immune system to target HPV16-positive solid tumors. **Nivolumab** is a monoclonal antibody immunotherapy that acts as a programmed death-1 (PD-1) immune checkpoint inhibitor, while **ipilimumab** is a monoclonal antibody targeting cytotoxic T-lymphocyte-associated protein 4 (CTLA-4), another checkpoint inhibitor. The rationale behind this combination is to enhance anti-tumor immune responses by both priming (via SQZ-AAC-HPV) and releasing immune checkpoints (via nivolumab and ipilimumab) for the treatment of patients with advanced or metastatic HPV16-positive solid tumors. The regimen is under evaluation in early-phase clinical trials for safety and preliminary efficacy[1][3][5].
