SQZ-PBMC-HPV + atezolizumab is a combination immunotherapy regimen consisting of an autologous antigen-presenting cell vaccine (SQZ-PBMC-HPV) and an immune checkpoint inhibitor (atezolizumab). SQZ-PBMC-HPV is manufactured by loading a patient's own peripheral blood mononuclear cells (PBMCs) with human papillomavirus 16 (HPV16) E6 and E7 antigens using microfluidic "Cell Squeeze" technology. This enables efficient MHC-I presentation of HPV16 antigens and robust activation of HPV16-specific CD8+ T cell responses. Atezolizumab is a monoclonal antibody that inhibits Programmed death-ligand 1 (PD-L1), an immune checkpoint, thereby enhancing T cell-mediated antitumor immunity. This combination is under investigation in patients with advanced or metastatic HPV16-positive solid tumors, including anal, cervical, head and neck, penile, vulvar, and vaginal cancers. The rationale is to synergize enhanced antigen-specific T cell priming by SQZ-PBMC-HPV with checkpoint blockade by atezolizumab to overcome immune evasion and maximize antitumor effects[1][3][4][5].
