Overview

SR-14968 is a small molecule, G protein-biased agonist of the mu-opioid receptor (MOR) belonging to the SR compound family. Characterized by a piperidine core structure, it was developed at The Scripps Research Institute to selectively activate G protein signaling pathways while minimizing the recruitment of β-arrestin2. This functional selectivity is intended to decouple analgesic efficacy from typical opioid-related adverse effects, such as respiratory depression and gastrointestinal dysfunction. Preclinical studies in murine models demonstrate that SR-14968 provides potent, long-lasting antinociception and a wide therapeutic window. However, research also indicates that the compound possesses significant addictive potential, including rewarding properties in conditioned place preference tests and the induction of robust physical dependence and naloxone-precipitated withdrawal symptoms.
