SR12343 is a **small molecule inhibitor** that acts as a mimetic of the NEMO-binding domain (NBD), specifically disrupting the interaction between IKKβ (IκB kinase β) and NEMO (NF-κB essential modulator, also known as IKKγ) within the canonical NF-κB pathway[1][2][3][4][8][10]. This selective inhibition blocks TNF-α- and LPS-induced NF-κB activation, reducing downstream inflammatory signaling and the senescence-associated secretory phenotype (SASP)[1][2][3][7][10]. Preclinical studies show it can suppress acute pulmonary inflammation and fibrosis, reduce cellular senescence, and attenuate muscle wasting and other degenerative pathologies in models of aging, Duchenne muscular dystrophy, and chemotherapy-induced cachexia[6][7][10]. SR12343 is currently for research use only and not approved for clinical application[1][2][8].
