SR29065 is a small molecule agonist of the nuclear receptor REV-ERBα (NR1D1), a critical component of the mammalian circadian clock. Developed by researchers at Scripps Research, SR29065 is used as a pharmacological tool to investigate the role of circadian signaling in inflammation, metabolism, and cellular homeostasis. In the context of respiratory research, SR29065 has been shown to mitigate cadmium-induced injury in alveolar type 2 (AT2) epithelial cells. By activating REV-ERBα, the compound helps restore epithelial homeostasis, reduces the expression of mesenchymal and profibrotic markers (such as vimentin and α-SMA), and inhibits apoptosis. This suggests a potential therapeutic role for REV-ERBα agonists in treating chronic lung diseases like chronic obstructive pulmonary disease (COPD) and pulmonary fibrosis, where environmental toxicants disrupt alveolar integrity.
