SRA737 + gemcitabine + cisplatin is an experimental combination therapy under investigation for advanced solid tumors. SRA737 is an orally bioavailable, potent and highly selective small molecule inhibitor of checkpoint kinase 1 (Chk1), a key regulator of cell cycle progression and the DNA Damage Response (DDR) pathway[1][2][5][6][8]. SRA737 induces synthetic lethality in cancer cells with elevated replication stress, often due to oncogenes or DNA repair deficiencies[1][5]. Gemcitabine is a nucleoside analog used as a chemotherapy agent, interfering with DNA synthesis, while cisplatin is a platinum-based chemotherapy that causes cross-linking of DNA and leads to apoptosis. In combination, these drugs aim for synergistic antitumor effects: SRA737 impairs DNA repair, gemcitabine further induces replication stress and DNA damage, and cisplatin introduces DNA crosslinks, resulting in enhanced cancer cell death, particularly in tumors with genetic features linked to high replication stress[1][2][5][6]. This combination is being studied primarily in genomically selected advanced cancers, including anogenital cancer, cervical cancer, high-grade serous ovarian cancer, rectal cancer, and small cell lung cancer[2][4][6].
