SRB17 is a best-in-class, fully human IgG monoclonal antibody targeting the Human Epidermal Growth Factor Receptor 3 (HER3/ErbB3). Developed by SunRock Biopharma in collaboration with the University of Stuttgart, SRB17 exhibits high affinity (picomolar EC50) and a unique mechanism of action by binding to HER3 and maintaining it in a closed conformation. This prevents HER3 from dimerizing with other receptors such as HER2 or EGFR, thereby interrupting downstream oncogenic signaling even in the absence of its ligand (neuregulin). The antibody is being developed for the treatment of HER3-overexpressing malignancies, including head and neck cancer, HER2-amplified breast cancer, and metastatic colorectal cancer. SunRock Biopharma has also partnered with Debiopharm to utilize SRB17 in the development of HER3-targeting antibody-drug conjugates (ADCs).
