SRB19 is a tetravalent bispecific antibody targeting both Human Epidermal growth factor Receptor 3 (HER3) and Epidermal Growth Factor Receptor (EGFR). Developed by SunRock Biopharma, it is engineered as a single-chain diabody-Fc fusion protein. SRB19 is designed to simultaneously inhibit both ligand-dependent and ligand-independent phosphorylation of EGFR and HER3, which are frequently co-expressed in various solid tumors and contribute to treatment resistance. Preclinical studies have demonstrated that SRB19 can induce tumor regression and extend survival in models of head and neck squamous cell carcinoma (HNSCC) and triple-negative breast cancer (TNBC). Additionally, it has shown synergistic effects when combined with cetuximab. The drug is currently in preclinical development and has been licensed to Debiopharm for further advancement, including potential development as an antibody-drug conjugate (ADC).
