SRI-011381 is a small molecule agonist of the transforming growth factor-beta (TGF-β) signaling pathway, specifically acting as an activator of the TGF-β receptor type I (ALK5) and the downstream SMAD2/3 signaling cascade. It is extensively utilized in preclinical research as a pharmacological tool to stimulate TGF-β signaling and investigate its role in diverse pathological processes. Studies have demonstrated its potential in promoting amyloid-beta clearance in Alzheimer's disease models by enhancing TMEM119 expression, alleviating symptoms of opioid withdrawal, and mitigating various forms of organ fibrosis, including pulmonary and myocardial fibrosis. Furthermore, SRI-011381 has been shown to support the self-renewal of human pluripotent stem cells and protect against oxidative stress-induced damage in oocytes and gastric mucosa.
