Drug intelligence / Profile preview
SRI-42718 is a first-in-class, orally available, non-nucleoside small molecule antiviral agent that targets the conserved non-structural protein 4 RNA-dependent RNA polymerase (nsP4-RdRp) of alphaviruses, including Chikungunya virus (CHIKV). Developed by Oregon Health & Science University in collaboration with Southern Research and the University of North Carolina School of Medicine, SRI-42718 blocks both genomic RNA (gRNA) and subgenomic mRNA (sgmRNA) synthesis, as well as viral protein production. In preclinical models of CHIKV infection, oral administration of SRI-42718 significantly reduces acute viremia, viral tissue burden, and virus-induced joint swelling, and also decreases viral RNA levels in joint-associated tissues during the persistent phase of infection.
