Drug intelligence / Profile preview
SRI-43887 (also referred to as duali) is a novel small-molecule dual inhibitor of receptor-interacting serine/threonine-protein kinase 3 (RIPK3) and bromodomain-containing protein 4 (BRD4). Developed by Southern Research and the University of Alabama at Birmingham, the compound is designed to target necroptotic and inflammatory pathways that are upregulated following exposure to arsenical chemical warfare agents, such as lewisite. In preclinical models of arsenical-induced acute kidney injury (AKI) and skin injury, SRI-43887 has demonstrated protective efficacy, including the recovery of glomerular filtration rate and reduction of inflammatory markers like interleukin-6 (IL-6).
