SRI-7330 is a small molecule inhibitor of thioredoxin-interacting protein (TXNIP), a protein that plays a central role in regulating cellular redox balance, the unfolded protein response (UPR), and autophagy. TXNIP is frequently overexpressed in multiple myeloma (MM) cells compared to normal plasma cells, particularly in patients with 1q amplification. SRI-7330 has demonstrated the ability to reduce TXNIP expression, induce cell death, and activate the apoptotic arm of the UPR, as evidenced by the increased expression of pro-apoptotic markers such as CHOP and GADD34. Research suggests that TXNIP inhibition via SRI-7330 may be a viable therapeutic strategy for hematological malignancies, especially when combined with proteasome inhibitors or CREB1 inhibitors.
